Oxidative phosphorylation in fractionated bacterial systems. XXII. The effect of near ultraviolet irradiation on the succinate oxidase pathway of Mycobacterium phlei.
نویسندگان
چکیده
The succinate oxidase activity of Mycobacterium phlei was sensitive to irradiation with light at 360 mp. The loss of activity was due to a lesion occurring in the electron transport chain between the flavoprotein and cytochrome b regions. The activity lost was not restored by addition of vitamin Kr or other cofactors but could be restored by the addition of the supernatant fraction. The factor responsible for the restoration of the succinate chain has been purified. It was sensitive to heat and irradiation and appeared to be protein in nature. It activated both oxidation and phosphorylation in irradiated particulate systems. The proteins which restored the two activities could not be separated.

Oxidative phosphorylation in fractionated bacterial systems. XXV. Studies on the involvement of metal in Mycobacterium phlei.
The involvement of metal in oxidation and coupled phosphorylation by cell-free preparations of Mycobacterium phlei was examined. The oxidation of succinate was inhibited by metal-chelating agents, but with /3-hydroxybutyrate, chelating agents exhibited an uncoupling effect. The site of inhibition was found to be between the flavoprotein and the cytochrome b regions of the electron transport cha...

Spectrophotometric evidence for the reduction of nonheme iron in the respiratory particles of Mycobacferium phlei was obtained by trapping the metal as ferrous o-phenanthrolinate. The protein-bound iron-o-phenanthroline complex had an absorption maximum at 510 mp, and the extent of reduction of nonheme iron was followed by observing the increase in absorbance at 510 to 540 rnp on the addition o...
متن کاملOxidative Phosphorylation in Fractionated Bacterial Systems XVIII. PHOSPHORYLATION COUPLED TO DIFFERENT SEGMENTS OF THE RESPIRATORY CHAINS OF MYCOBACTERIUM PHLEI*
A number of bacterial systems have been described which will carry out oxidative phosphorylat, ion (1-12). In general these systems differ from intact mammalian mitochondrial systems in that the bacterial systems have lower P:O ratios and fail to exhibit respiratory control. Loss of sites of phosphorylation (13, 14) and nonphosphorylative electron transport bypass reactions (13, 15-18) has been ...
